Description

Primary Objective:

To establish a robust process for the analysis of genomic alterations and influence of cytokines on tumorigenic potential and stem cell charateristics in primary colon cancer and ulcerative colitis specimens.

Secondary Objectives:
  1. To determine if gene defects acquired by xenografts and spheres from ulcerative colitis and colorectal cancer follow those in the classical adenoma-to-carcinoma pathway.
  2. To determine if spheres from colon cancer initiating cells are true cancer stem cells which mimic the clonal and metastatic behavior of human colorectal cancer.
  3. To show the key contribution of IL8 in ulcerative colitis stroma to epithelial proliferation.
  4. To define the effects of IL8 on the in vivo and in vitro formation of tumors derived from colon cancer initiating cells isolated from colitic colon.
  5. To determine the influence of downstream mediators of IL8 signaling on the initiation of tumors induced by colon cancer initiating cells derived from colitic colon.
  6. To delineate the contribution of gain-of-function p53 mutations to IL8-induced potentiation of tumorigenicity.
Inclusion Criteria

Inclusion Criteria

  1. Consenting subjects > 18 y/o presenting for operative intervention of their primary colorectal cancer or inflammatory bowel disease related-colitis or colitis-associated cancer.
  2. The participants will be identified from the PI's own patient cohort as well as from the Colorectal Surgery department at the Cleveland Clinic who are presenting for surgical resection of their disease.
  3. The subjects will be identified from the PI's practice and from the departmental practice as discussed preoperatively and identified by reviewing the operating room schedule.
  4. The subjects will be contacted by phone if not able to be consented in clinic. If written consent is not obtained in the preoperative setting then formal written consent will be obtained in the preoperative holding area if verbal consent was affirmative.
